Quinolimide-based peptide biosensor for probing p25 in vitro and in living cells

CDK5 kinase is activated through interactions with different partners including the p35/p25 regulators. Active plays a key role in several neuronal functions and its hyperactivity contributes to variety of neurodegenerative processes human cancers, particular glioblastomas neuroblastomas. In order probe that interact CDK5, we designed synthesized fluorescent quinolimide-labelled peptides derived from C-helix which were implemented vitro by cellulo imaging U87 glioblastoma N2a neuroblastoma cells. Ectopic expression NIR protein miRFP670 fusion p25 (p25-miRFP670) cells induced FRET between peptide biosensor p25-miRFP670, fluorescence intensity was proportional level p25, thereby demonstrating potential report on relative abundance.
